Application of LAMP Method to Distinguish Anguilla marmorata from A. bicolor pacifica

Loop-mediated isothermal amplification (LAMP) method was employed to identify Anguilla marmorata and A. bicolor pacifica. Partial sequences of mitochondrial DNA 16S rRNA were used to design LAMP primers that were specific to A. marmorata. To test the sensitivity of the LAMP method, different amounts of template DNA were used. It showed that 1 pg of the total template DNA could be detected by both electrophoresis and visual florescence. The result showed that A. marmorata could be easily distinguished from A. bicolor pacifica within an hour. This suggested that LAMP assay could be used to identify other species.
