Characterization and Identification of a Probiotic Bacterium, Micrococcus luteus

Morphological, physiological and biochemical characterizations were conducted for the identification of an isolated bacterium LK1012W. The results showed that the characters of LK1012W were Gram positive (+) coccus, Catalase (+), Oxidase (?), endospore (?), able to grow up on aerobic medium (+) and unable to grow on anaerobic medium (?), with DNA G + C content (16S rDNA) of 72.8 mol%. The LK1012W strain was similar to three species of Micrococcus: M. luteus,M. yunnanensis and M. endophyticus, based on 138 characteristics tested and 16s rDNA sequenced. The results of DNA hybridization showed that LK1012W vs M. luteus BCRC 11034 exhibited 89% similarity while that of LK1012W vs M. yunanensis BCRC80243 was 80%. Upon the above results, the LK1012W strain was therefore identified as M. luteus.
